Pyruvate dehydrogenase complex.

Hello my readers...
The most haunting topic for me in biochemistry is Kreb's Cycle. In fact for me it's more like crib cycle :p

Plus to add on the boring factor, we have to learn some of important enzymes and their mechanisms in depth.

Hence today we will be learning about the PDH complex

The combined dehydrogenation and decarboxylation of pyruvate to the acetyl group of acetyl-CoA requires the sequential action of three different enzymes and five different coenzymes or prosthetic
groups. They are:

Three enzymes include:

  1. Pyruvate dehydrogenase (E1)
  2. Dihydrolipoyl transacelylase  (E2) [ I always have hard time pronouncing it :( ]
  3. Dihydrolipoyl dehydrogenase (E3)

Five cofactors include:

  1. Thiamine phosphate (TPP).
  2. Flavin adenine dinucleotide (FAD)
  3. Nicotinamide adenine dinucleotide (NAD).
  4. Coenzyme A (CoA or CoASH).
  5. Lipoate.
Mnemonic:

Trust me, that's her beauty secret.

Anways , back to the subject

Now the mechanism of PDH cxomplex is right here:

Hope it helps you all.
Thanks a lot
