Differentiate between H and O antigen of Salmonella and how to remember it!

Today we will have a look at differences between two famous antigens of Salmonella.
So without wasting any time, let's get straight into it.

Let's start.

What type are both the antigens?
H antigen is the flagellar whereas O is a somatic antigen present on the cell wall.

Sensitivity?
H antigen is heat labile and sensitive to alcohol. O antigens are heat stable and remains unaffected by alcohol.

Reagents used for antigen extraction?
H antigen is extracted by treating the cells with formaldehyde while trichloroacetic acid is used for extraction of O antigens.

What is the composition of both the antigens?
H antigen is made up of a protein complex. O antigens are made up of a phospholipid - protein - polysaccharide complex.

What type is the agglutination reaction?
On agglutination, H antigen produces large, fluffy and loose clumps and O antigen produces compact, chalky and granular clumps.

Immunogenicity?
H antigen is highly immunogenic and induces antibody formation with high titres. O antigen is less immunogenic and produces antibody formation with low titres.
